TriMScope I - The Realtime 2-Photon Microscope

La Vision BioTec’s innovative approach to 2-photon microscopy is unique as the TriM Scope supplies various unique features.

 

First, the user can choose if the system should run in classical single beam scanning mode or in LaVision BioTec fast multi beam mode. The multi beam mode allows the user to run up to 64 beams simultaneously, which delivers unmatched image acquisition rates at minimum sample damage for 3 main reasons: 

 


First, 64 beams deliver 64 times more fluorescence light keeping the intensities in individual beams – and therewith the damage – low.

 

Second, more fluorescence light allows higher frame rates.

 

Third, CCD cameras deliver much better sensitivity than PMTs.


TriM Scope - Typical Scan Path

 

To deliver these results the TriM Scope utilizes a unique imaging setup that scans the specimen with 64 beams simultaneously. First, 64 beams deliver 64 times more fluorescence light keeping the intensities in individual beams – and therewith the damage – low. Next, more fluorescence light allows higher frame rates. Finally, CCD cameras deliver much better sensitivity than PMTs.

 




Next, the TriM Scope supports up to 8 NDD detectors for classical single beam microscopy applications, up to two CCD cameras and one TCSPC detector for FLIM measurements.  Up to 3 NDD detectors can be placed close to the objective lens and up to 2 NDD detectors can work in transmission. In addition NDD detectors can be placed on top of the fluorescence arm of an uptight microscope or at one baseport of an inverted microscope. All NDD detectors can be GaAsP or classical bialkali PMTs. 

 

Furthermore, LaVision BioTec´s innovative ImSpector Pro software supports advanced scan modes and allows the user to switch within different scan areas within xx ms. For this reason the TriM Scope is the perfect tool for actual application like photo activation, uncaging and FRAP.

 

Finally, the TriM Scope can be combined with an OPO. The OPO shifts the Ti:Sa wavelength from 800-880 nm to 1100-1300 nm. So red dyes or proteins can be exited effectively in 2-photon mode, which results in low bleaching rates and deepest penetrations depth. In LaVision BioTec’s setup the Ti:Sa laser and the OPO can be scanned simultaneously which allows to excite green/yellow and red dyes at once.

  • Various CCD cameras (single or dual camera detection) that deliver highest frame rates, low noise at maximum quantum efficiency
  • EOM for FRAP, Photo-Activation or Uncaging capabilites
  • Optional FRAP, Photo-Activation or Uncaging kits
  • Up to 8 NDD PMT-detectors for full flexibility in standard single beam operation
  • Up to 2 transmission NDD detectors for «SHG imaging» or improved collection effeiciency
  • Fast filter wheels and imaging spectrometers for spectral detection
  • The TCSPC detector for «FLIM -Fluorescence Life Time Microscopy»imaging
  • OPO kit for simultanious OPO and Ti:Sa scanning

 

All options are integrated into LaVision BioTec’s image acquisition and processing software package ImSpector.
