Integration into LaVision BioTec’s TriM Scope II
The microendoscope is designed as an upgrade for LaVision BioTec’s TriM Scope II. Due to the modular design, the user can easily switch between standard and endoscope mode without re-alignment.

360° Panorama View Endomicroscope
In vivo imaging with sub-cellular resolution is limited to a depth of 1 mm due to the tissue’s optical density, scattering and absorption of the emitted fluorescence light. 2-photon endomicroscopy as a minimal invasive approach enlarges the imaging volume by at least one order of magnitude by going deeper into the tissue and recording 360° panorama images! Moreover, spiral panorama images of e.g. the striatum deep in the mouse brain can be recorded.
The flexible design enables the implementation into LaVision BioTec’s TriM Scope II but also in existing standard two-photon microscopes from other manufacturers.

Integration into further 2-photon microscopes
Alternatively, a mechanical kit allows the endoscope device to be mounted directly on the optical table enabling implementation into laser-scanning microscopes from other manufacturers.
Principle of operation
The Endomicroscope optics consist of a standard laser-scanning objective lens and an additional rod lens in the tip. A prism at the tip deflects light by 90° and therefore allows side viewing. Furthermore, motorized rotation of the rod lens allows 360° panorama view dataset recording deep inside live tissue. The whole Endomicroscope can be moved along the microscope’s optical axis by a linear precision drive. This allows to change the depth within the tissue and recording of spiral panorama datasets.
A) 360° panorama view imaging mode
At a selected depth within the tissue motorized rotation of the rod lens enables 360° panorama view 2D or 3D recordings.
B) 3D volume acquisition mode
A 2D image is recorded by scanning the endoscope focus with the scanner technology. Motorized changing of the focus in front of the endoscope allows it to address consecutive z planes in order to record a 3D dataset.
C) 360° spiral panorama view mode
By consecutive z-movement of the whole endoscope in combination with rotation of the rod lens, 360° Spiral Panorama 2D or 3D datasets can be recorded.
