Sample motion due to heartbeat and breathing makes in vivo microscopy difficult leading to blurred image or losing sight of the area of interest.

LaVision BioTec's Approach
LaVision BioTec’s in vivo stabilization package comes with
- Piezo driven Tripod moving your objective in all dimensions
- Additional software based correction (x and y)
The Tripod is driven by innovative software algorithms calculating the movement of an object, e.g. a neuron, during data acquisition. The compensation runs up to 250 frames/s.

Figure 1: 2-photon in vivo calcium imaging of an awake mouse using GCaMP as calcium indicator. Ca2+ signals were recorded with a resonant scanning microscope (TriMScope II, LaVision BioTec) while the mouse was running on a treadmill. The images are snapshots of two superimposed sequences. Without motion stabilization (light blue), the neuron moves around the center of the screen. By using LaVision BioTec’s in vivo stabilization package, lateral and axial shifts are compensated and the neuron is kept in the center over time (red).
Figure 2: Fluorescence signal over time with (red) and without (light blue) motion stabilization. By using LaVision BioTec’s in vivo stabilization package, the noise of the baseline can be minimized making Ca2+ signals detectable.
Connections and Dimensions
The in vivo stabilization package comprises the tripod (including the spacer), an electronic box and a plug-in for ImSpector for direct control by PC.
